چکیده

This paper presents a PC-based PCR thermal cycler with a host-local architecture, where a PC is employed as the host. The local system only executes the thermal cycling process and the PC is responsible for the management of PCR protocols with a convenient GUI. The presented host-local system greatly reduces the development time and the maintenance cost, providing an economical PCR thermal cycler.

Interlaboratory study on thermal cycler performance in controlled PCR and random amplified polymorphic DNA analyses.

BACKGROUND Intercomparisons of PCR-based data between laboratories require an assurance of assay reproducibility. We performed an interlaboratory study to investigate the contribution made by a variety of thermal cyclers to PCR performance as measured by interblock reproducibility and intrablock repeatability. METHODS Two standardized assays designed to minimize the introduction of non-therma...

A Rapid and Low-Cost PCR Thermal Cycler for Low Resource Settings

BACKGROUND Many modern molecular diagnostic assays targeting nucleic acids are typically confined to developed countries or to the national reference laboratories of developing-world countries. The ability to make technologies for the rapid diagnosis of infectious diseases broadly available in a portable, low-cost format would mark a revolutionary step forward in global health. Many molecular a...

Rapid microfluidic thermal cycler for polymerase chain reaction nucleic acid amplification

Polymerase chain reaction (PCR) is widely used in biochemical analysis to amplify DNA and RNA in vitro. The PCR process is highly temperature sensitive, and thermal management has an important role in PCR operation in reaching the required temperature set points at each step of the process. The goal of this research is to achieve a thermal technique to rapidly increase the heating/cooling therm...

Optimisation of DNA and RNA extraction from archival formalin-fixed tissue.

Archival, formalin-fixed, paraffin-embedded tissue is an invaluable resource for molecular genetic studies but the extraction of high quality nucleic acid may be problematic. We have optimised DNA extraction by comparing 10 protocols, including a commercially available kit and a novel method that utilises a thermal cycler. The thermal cycler and Chelex-100 extraction method yielded DNA capable ...
